Nato a Hollywood il 5 marzo 1936 da genitori già nell'ambiente, gli attori Nina Olivette e Harry Stockwell (la voce del "Principe Azzurro nel disneyano Biancaneve e i sette nani"), presto Dean si unisce al fratello più grande, Guy, diventando anch'egli un attore bambino. E' molto richiesto grazie alla sua aria innocente e alla graziosa faccia da cherubino, coronata di riccioli. A 21 anni è già apparso in 21 film.

A 16 anni lascia Hollywood e inizia a viaggiare per l'America facendo vari lavori. Torna a recitare a New York, in una produzione di Broadway, "Compulsion", ruolo che ha ripetuto nel film del 1959 (che gli vale un premio al Festival di Cannes).
Solidifica la sua reputazione con la matura interpretazione di un giovane inquieto in Sons and Lovers (1960), e riceve un secondo premio a Cannes per il suo ruolo in Long Day's Journey Into Night (1962). Ma Stockwell smette di recitare ancora una volta.

Negli anni '70 appare solo in qualche piccola parte, con il suo amico Dennis Hopper, ma è negli anni '80 che compie uno spettacolare ritorno con importanti ruoli di supporto in "Dune", "Paris, Texas" (entrambi 1984), "Blue Velvet" (1986), "Gardens of Stone" (1987), e un cameo come Howard Hughes in "Tucker: The Man and His Dream" (1988); ritorno che culmina in una nomination all'Oscar per il suo riuscito passaggio al comico come boss da quattro soldi in "Married to the Mob" (1988), e lo porta ad essere scelto per la serie tv "Quantum Leap" (1989-93). Appare anche in "Limit Up" (1989) e nel divertente ruolo di disperato sceneggiatore in "The Player" di Robert Altman (1992). Sono questi ruoli da attore di mezz'età che gli hanno portato maggior successo e popolarità.

Uno dei pochi bambini prodigio riuscito nel continuare a recitare con successo anche da adulto, Stockwell è apparso in innumerevoli film ed ha ricevuto varie nomination e premi sia per i suoi lavori televisivi che per quelli cinematrografici, tra cui un Golden Globe per quello che è probabilmente il suo ruolo più famoso, l'osservatore olografico Albert Calavicci in "Quantum Leap."
